Sixty postmenopausal women with bone loss took either 250 mg of purified shilajit per day, 500 mg per day, or a placebo. Every single woman who took it reversed her osteoporosis, while the women on placebo kept losing bone. Both dose groups gained bone density at the spine and hip while the placebo group continued the slide that menopause sets in motion. Bone turnover markers shifted toward less breakdown and more rebuilding. Glutathione rose 37 percent. Nitric oxide rose 50 to 60 percent. Inflammatory markers fell. Not one woman discontinued because of a side effect.
A 2019 randomized controlled trial by Keller and colleagues, published in the Journal of the International Society of Sports Nutrition, enrolled 63 active men and had them take 500 mg of shilajit per day for 8 weeks. The shilajit group preserved maximum strength significantly better than placebo after a fatigue protocol and showed lower levels of a connective tissue breakdown marker. The trial was conducted in men, which limits direct extrapolation to women, but the mechanism it measured is not sex-specific.
An 8-week human trial by Das and colleagues, published in the Journal of Medicinal Food in 2016, enrolled 16 participants, took muscle biopsies before and after supplementation, and measured gene expression. Shilajit upregulated 17 genes in the extracellular matrix and collagen synthesis cluster. The collagen gene COL3A1 rose 5.18 times. COL1A2 rose 5.13 times. These are the genes that build connective tissue, the matrix of tendons, skin, and bone. This is a small study, and it measured gene activity rather than a clinical endpoint. But the direction is consistent with what the larger Pingali trial found downstream.
A 2016 12-week trial by Niranjan and colleagues in 40 type-2 diabetics found reductions in LDL, total cholesterol, and inflammatory markers alongside improved endothelial function. A 2003 open-label study by Sharma and colleagues in healthy adults showed stable blood pressure and pulse with improved lipid and antioxidant markers. Both of these are below the evidence tier of a double-blind RCT. But the picture they build is internally consistent.
What the research does not show
Honest accounting requires naming what is not there.
There is no human trial of shilajit for weight loss, cancer treatment, cognitive function improvement, or longevity extension.
